This line associates (or rather compares) wealth and illness and even transforms the idea of possession into one of â€Å"being possessed.† In this way, the wealth that Volpone has accrued actually possesses him, and the illness that he has been feigning from the very beginning of the play in his effort to obtain wealth seems to possess Volpone as well. Volpone was never actually â€Å"ill,† but by this new definition Volpone was actually ill this entire time – sick with wealth, thus making wealth a disease. In fact, all the characters in this play are so infatuated with wealth that by definition they all would be sic k. The introduction to this edition of Volpone mentions that the Christian concept of demonic possession is present in this play as well, but â€Å"what really possesses all of [the characters] is greed for gold† (Jonson 11). Perhaps what is also most interesting is one of Volpone’s earlier lines when he says â€Å"I rather pity their folly and indiscretion than their loss of time and money, for those may be recovered by industry, but to be a fool born is a disease incurable† (Jonson 95, emphasis mine). Regarding previously discussed notions, Volpone seems to be the most diseased of all, yet he is the one giving the definition of who is diseased. Thinking back upon Jonson, this is a very interesting (and ingenious) way to critique wealth and the search for it. As aforementioned, Jonson came from the lower class, and it seems that he is making some serious critique in this line regarding who is diseased and who is not. One cannot help the class that they are born into per se, but morals and character are things that a person has much agency over. Being born a â€Å"fool† here, when Jonson’s very voiced moral intentions are taken into account, seems to mean a person born believing that possession, wealth, and power are the only valuable things in the world is the true fool instead of the person born into poverty. In this way, it’s almost as if Volpone is unaware that he is admitting he is the fool since he essentially worships wealth, possession, and power – and he is thus diseased both falsely through his feigned disease and literally through his obsession with wealth. Linking this to the notion of power, it seems as if illness is inextricably (and perhaps a bit difficultly) associated with wealth and power. As previously discussed, there is a tangled (but brilliant) relationship between wealth and possession where wealth is the center of the world and the ability to obtain it through the power of cunningness is also where much of its value lies. If an individual is able to â€Å"possess† wealth or people, then they most likely have the power of cunningness to obtain this wealth. In short, if a person possesses things, they will have power. However, with the line â€Å"these possess wealth as sick men possess fevers,† the connection between power, possession, and illness has come full circle (Jonson 206). By being wealthy, the characters are sick, but this seems to imply that the more wealth a person obtains (through the power of cunningness), the sicker a person becomes. This explains many of the â€Å"power switches† that occ ur throughout the play as in Act 3 where Volpone and Mosca exchange power as well as in Act 4 when Voltore seems to have the most power. All of the characters are cunning as well as seeking greedily for wealth, and because power, possession, and disease have been fused into essentially a single entity, all of the characters are sick – which allows these shifts to happen. To push this even further, the acting done by the characters (or rather actors) ties up these concepts even more. To a large degree, the â€Å"real† Volpone is never portrayed in this play because he is always so busy pretending to be something or someone that is he not. In a strange sense, it seems that whoever Volpone pretends to be, he becomes. Interestingly, the only time Volpone seems to be actually Volpone is when he is talking to Mosca in the first act and explicitly states his own name. This is really the only time that Volpone’s name is mentioned, and it essentially one of the only times where Volpone is actually not pretending to be something he is not (Jonson 77). With this in mind in regards to all that has already been discussed, it seems very plausible that Volpone is pretending to be something he is. He can only be Volpone in that brief moment when he declares that he is Volpone, but all throughout the rest of the play he is not Volpone, but in not being Volpone he is very much being Volpone (if that makes sense). In the beginning Volpone pretends to be dying, but since the real Volpone is never significantly portrayed, it’s almost as if he was never alive to begin with. Similarly, Volpone feigned sickness in the beginning, and he even mentions that â€Å"first I feigned diseases; now I have one† about Lady Politic (Jonson 121). Regarding the previous discussion about power, possession, and illness, it would then seem that Volpone was never feigning illness at all because his wealth was an illness therefore he was actually pretending to be something that he perpetually is – diseased through wealth. Additionally, Volpone’s â€Å"acting and the power this gives him over other people excites Volpone more than either gold or sexual possession† as mentioned in the introduction (Jonson 13). Volpone’s acting has now been equated to power, and by default has then been associated to disease and corr uption – Volpone’s illness was never fake, rather it was simply not the illness he thought it was. Strangely, in his pretending, he fooled even himself because being the actual â€Å"fool born,† he never realized that his wealth was his disease. Though it’s all rather tangled up into what might perceived as a congealed conceptual mess, there is indeed a tight relationship going on between power, possession, and disease, as well as acting/pretending. They are so inextricably linked that it seems almost impossible to sift through them individually to pull them apart, which is frustrating while also being brilliant that they are equated with each other so masterfully and exploited so horrifically that it’s impossible not to think about Jonson’s claims about providing morally based plays (or poems) that punish vice. Linking back to the tensions between Jonson and ownership as well as being a â€Å"good man† in order to be a â€Å"good poet,† it does seem that, though Jonson was far from perfect, intrinsically he could be a good man – this being emphasized through his intentions regarding his work – and perhaps his creations were the result of him trying to improve his public portr ayal (or even himself) as well as provide a moral lesson for the betterment of society. Methods: The authors took a qualitative design to their research to developed an understanding why the quality of care varied across several nursing homes (Burns, D. J., Hyde, P. J., Killett, A. This study included 12 different nursing homes which consist of a mixture of non-profit and for profit nursing homes and residential living communities over the course of three years (2009-2012). Each nursing home varied from 10 to 65 bed facilities that varied in the terms of the level of care provided. By looking at the poem in depth, I have come to a conclusion that Carol Ann Duffy is the persona who is writing with a childs perspective as when she was a child. In verse one, The poet creates a sense of noise and chaos by saying how each member of her family are making noise and moving. Mother singing My brothers cried both of these quotes contribute to the feeling of noise and chaos. Two metaphors, which are found in the poem by Carol Ann Duffy, could be fell through the fields miles rushed back. They show that they were in the countryside. It also shows that they are using some form of transport, which may be train or car. I assumed by reading the poem that the reader was a young girl, and when she is sitting down where on the journey, it feels like you are stationary so it feels as if everything is rushing towards yourself. This demonstrates the Childs prospective. On the last line of the first verse, the metaphor shows that the young girl is looking at one of her toys, which maybe a bear with button eyes, and as she has grown up a fraction more than she was, she may now understand that toys do not have feelings, and they are only buttons as the eyes so they cannot really see things. She makes us feel like a child is commenting here because when youre at such a young age, you think that toys really do live on their own world and they have the same senses as us. I. e. touch, taste, feel, hear, pain etc. I think that the whole poem focuses mainly on sudden interruptions because from each paragraph is a different age section of her life. You can also see by the poem that as each paragraph develops, the way it is written and the style of wording develops, as if the child is becoming increasingly mature and adult-like. The Childs vulnerability is shown in verse two by using such words as big boys eating worms and shouting words you didnt understand perhaps this is hinting that maybe the big boys who were eating worms were swearing or saying words which she was too young to have come across. The last word in verse two is italicised because the narrator (who is a daughter) is speaking and it also shows that she is missing where she used to live as a child. In verse three, the child is less vulnerable now she has settled in because she is now used to everything. But then you forget this shows that now the narrator has lived for quite a few years, she tends to forget her past easier. The narrator now speaks in a comforted tone and gives away the idea of being a lot more worried about life in general as compared to when she was younger and slightly unsure about moving. Now that she has fitted in well, she now understands how it feels like to be part of their community. The change is compared to the shedding of a snakes skin. They are compared with each other because it is in conjunction with her voice changing i. e. maybe she has changed language or they use a different accent. Or simply her voice could have just changed with age. Now that she is used to the changes, she feels as if she has completely lost her old voice I lost a river, culture, speech, sense of first space This could potentially be a good or bad thing but we assume it is for good because she prefers being accepted and fitting in when she didnt think she would ever. The person created in the poem, we could comfortably say that it is a girl, even though no evidence backs up this statement. Yeast Two-Hybrid Essay -- Biology, DNA

The initial transformation task required two yeast strains (both as Mav203; Leu–, Trp–, His–), with one containing the pDBLeuDa4 bait plasmid, and the other with just the pDBLeu plasmid. The two strains were each transformed with a pPC86DMID1(Y) prey plasmid, where (Y) referred to a full-length positive control (MID1) , a DBB strain (MID1DBB), and a DCC strain (MID1DCC). The transformation process with the two strains and three prey plasmid DNAs resulted in 6 (2x3) transformation mixtures (Table 1). The transformation mixtures were then spread-plated at 1x and 1/10x dilutions using half of each plate on an SC (synthetic complete) yeast growth medium minus selected amino acid(s); one plate without Leucine (SC-Leu), and two plates without Leucine and Tryptophan (SC-Leu-Trp). Plates were then incubated for 4 days at 30 ° C, however due to failed growth in the transformation plates for pPC86DMID1 (both yeast strains), and an additional set of results had to be obtained. The spread-plated results for each strain, transformation, result source, and their associated control cases can be seen in Table 2, with a â€Å"Yes† growth observation referring to confluent growth and â€Å"No† for non-confluent growth. Following the incubation period, each of the 6 transformation plates were sampled and underwent 4x 10-fold serial dilutions, which were then spot-tested to either an SC-Leu-Trp, or SC-Leu-Trp-His (minus Histidine) medium. Each strain-transformation occupied half of each plate, with a spot per dilution level (Figure 1). An additional set of spot-tests were also performed to test for auto-activation of the a4 bait protein, using a strain transformed with an unmodified pPC86 plasmid (MaV203+pDBLeuDa4+pPC86). Following another round of in... ...the DCC variant (for all spot dilution levels). The lack of growth for the DBB variant suggested the B-box region was a requirement for interaction/binding with the a4 protein, while the coiled-coil domain didn’t appear to be a requirement. Its removal however did result in noticeably decreased growth rates when compared to the full-length control test, suggesting its removed resulted in some level of decreased interaction with a4, whether it usually stabilised the binding process, or is removal disrupted the overall form and binding site is unknown, and would require additional testing. However given the external result source, it could have been that group’s sampling and dilution methods had resulted in decreased cell viability, though a simple retest could rule this out, and would be a good starting point to determine any interactions with the coiled-coil domain.

These last five weeks in EXP 105 has really helped me learn a lot about learning in general, and also it has helped me to understand myself as a learner. If I had to explain the concept of learning to someone who has not yet taken this class, I would tell them that it is simply knowing who you are as learner and knowing what are your intentions are when it comes to learning. Intentions alone play a big part in learning because that is the ultimate goal; becoming an intentional learner. An intentional learner knows how to approach learning by developing skills to read, write, and think critically. I always felt that learning itself was a full time job, but this class changed that view by teaching me how to learn, and also what my weak and strong points were. This helps you to manage time, and learning no longer feels like it takes forever. In this class we were given a learning connections inventory (LCI). It was a short survey to help us identify our learning patterns. My LCI scores were as follows: Sequence 27, Precision 28. Technical Reasoning 20, and Confluence 17. Sequence and precision are my two use first learning patterns. I use these patterns on the regular. I am a very organized person and I like to make sure that I have all the information that is necessary before I perform a task. I use technical reasoning as needed, and then I avoid Confluence. Confluence requires a lot of creativity and that is the area I lack in the most so I try to stay away from it as much as I can. No matter how we use our learning patterns though, it always comes a time where we must use them all whether we realize it or not. Sometimes it is necessary to combine all of the learning patterns to get a full understanding of what is happening, or supposed to happen. The benefits of knowing your unique learning patterns are for one you already know how you should approach different tasks to get them done. It takes the guesswork out and saves you time to go back and finish something you did not  understand later. While learning patterns are strong tools to have in your tool box by themselves, sometimes it is necessary to have strategies to go along with them. When it comes to sequence I do not always think that a strategy is necessary because it is always important for things to be organized for them to look neat and flow smoothly when it comes too writing at least, and that is how I tried to think about every learning pattern and assignment in this class. My strategy for mu use first precision is to keep it at a minimum without being boring, less really is more sometimes. Technical reasoning will remain as it is since it is use as needed. I definitely know that my strategy for confluence was to intensify it a little more so that I can try to warm up to it and try to incorporate it into more of my assignments. The way that I took advantage of my strengths in this course was actually using them when I was completing assignments. The biggest challenge of my learning patterns was figuring out when and how to use them. It gradually got better once we discussed FITing and decoding assignments. It did not take long for me to grasp those concepts and after that everything became clear and it all made sense on hoe it all fit together. That solved the problem of the learning patterns being hard to understand when it came to using them to complete assignments in the course. I didn’t get to use the FITing concept until the end, but I wish I would have known about it earlier. I definitely plan to use it in my future courses because it makes everything easier to understand and I enjoyed doing it. I didn’t think of it as extra unnecessary work. It was more like a map to me to tell me what I should do. In the future I will also use more of the library and student resources to help me with FITing. The thing that I enjoyed the most about this class w being given the opportunity to learn about myself and who I am as a learner. I definitely feel different about learning and I plan to use everything I learned in this course in future courses. My favorite assignments to do in this class were the LCI and then the FITing and decoding assignment. To make the class a little better and more fun I think at least another variation of the LCI should be included. I had taken online courses before so all of my expectations were met. I enjoyed this class overall.
